Metabolites profiling of plant secondary compounds

Objectives

This practical short course intends to provide an overview of the different steps needed to profile secondary metabolites in plants. The entire course will take place in the laboratory where participants will have the opportunity to become familiar with sample preparation, analysis by LC-MS, GC-MS and NMR, and compound identification.

Content and pre-course assignements

The alkaloid-containing plant Atropa belladonna L. (nightshade) will be used as a case study with the aim to selectively extract and profile the alkaloids present in its tissues.

  • Selection of an appropriate solvent for alkaloid extraction
  • Enrichment of the alkaloid extract
  • Analysis by LC-MS, GC-MS and NMR
  • Identification of alkaloids
  • Discussion

Before the course, participants are requested to:

  • scan the literature about how to prepare and analyse alkaloids from plant tissues
  • establish a list of the different alkaloids present in nightshade

During the course, participants must wear long pants and closed shoes.

Lab coats and glasses must be worn during the course, and will not be provided (participants must bring their own material).

  • This course is free and open to all PhD students, however until 15 August priority is given to "Interuniversity doctoral program in organismal biology" participants.
  • Post-docs are welcome as long as places are available.
  • Maximum number of participants: 12 people (minimum 6 participants).
